Introduction

 First of all, we would like to express our deepest sympathies to all those affected by the Noto Peninsula earthquake that occurred on January 1. We sincerely hope for your safety and a speedy recovery.

 Since last year, in addition to the political fund party issue in Japan, there have been a series of events in the world of entertainment that have exposed the old pus. Looking at the world, the ongoing conflict between Ukraine and Russia, the advance of Israeli forces into the Gaza Strip in Palestine, and the attack by U.S. and British forces on a militia stronghold in Yemen, all remind us of the turbulent and confusing times we are living through. On the other hand, there is also bright news that reminds us of a new era in a positive sense. In the field of science, last year the NIH updated the reference sequence of the human genome, the PanGenome, for the first time in about 20 years. Compared to the previous reference sequence, the new sequence has much greater racial diversity, and is expected to be very useful for the development of personalized medicine in the future. In addition, the use and development of generative AI, such as ChatGPT, progressed dramatically last year. Technology (semantic decoder) has been developed that can recover words, phrases, and sentences from brain activity while listening to audio, and can also recover sentences that explain the content of a scene from brain activity while watching a silent video.

 The year 2024 is the year of the Chinese zodiac sign “Ki-no-Etsu” (甲辰). The “Kou” in the Chinese zodiac sign of “Koujin” is the first of the ten signs of the Chinese zodiac and has the meaning of the beginning or growth of things. For us pharmacokinetic researchers, I hope that this year will also be a year in which we will be able to produce a succession of groundbreaking research results and ideas that will be of real value to patients suffering from diseases, in conjunction with the development of cutting-edge science and technology!

 At the General Assembly of Delegates held last November, the 18th term officers including President Masao Kato and Vice President Hiroyuki Kusuhara were inaugurated, and the Newsletter Committee has started its activities under a new structure, including the replacement of some members. We will continue to make efforts to produce attractive contents of the Newsletter as a place for information exchange among members and information dissemination from members so that we can contribute to the further development of pharmacokinetics research, even if only slightly.  My name is Takato Tomabechi, Department of Pharmacology, School of Pharmaceutical Sciences, Kitasato University. I would like to thank the editorial board members and everyone involved for the opportunity to contribute to the newsletter of the Japanese Pharmacokinetics Society. I entered the Faculty of Pharmaceutical Sciences, Tokyo University of Pharmacy and Life Sciences in 2013 and started my research career in the Department of Pharmacokinetics and Pharmacodynamics led by Professor Katsuo Inoue in my fourth year. In April of the same year, I joined the Department of Pharmacology, School of Pharmaceutical Sciences, Kitasato University, led by Dr. Kazuya Maeda, and have been entrusted with research and, to a lesser extent, lecturing. I am about to enter my ninth year of teaching (though it has been less than a year since I started teaching).

 Good afternoon! Since the last issue, I have been writing about “Introduction to Antibody Drug Conjugate (ADC) Pharmacokinetic Studies”. In this third article, I would like to talk about bioanalysis (analysis of drug concentration in biological samples) of ADCs.

 First, I would like to talk about the subject of analysis. In order to understand the pharmacokinetics of ADCs with complex molecular structures, it is necessary to analyze each component derived from the molecular structure. The Draft Guidance (1) for clinical pharmacology studies of ADCs issued by the FDA in February 2022 recommends that ADCs with at least one payload, total antibody as the sum of all off-payloaded naked antibodies and ADCs, quantitation of the free payload and its active metabolites, and anti-drug antibody (ADA) evaluation be performed. … ( read more on the NL website / members only )
